Research Interests
My research is focused on four areas: 1.) clinical studies in patients with GI cancers (principally colorectal cancer); 2.) translational studies done on biologic specimens from patients with cancer done with laboratory collaborators; 3.) development of new cancer drugs and drug combinations; and, 4.) clinical trials methodology.

I am the Principal Investigator of national intergroup study N9741, an international Phase III study of oxaliplatin, irinotecan and 5-fluorouracil in combinations for first-line treatment of advanced colorectal cancer. New drug combinations are tested with the endpoints of time to progression, response rate, survival, toxicity and blood samples are examined for pharmacogenetic correlations to toxicity. This trial has been a major factor in changing the standard of practice for treatment of metastatic colorectal cancer in North America. It has also pioneered the field of pharmacogenetics in determining both treatment response and toxicity in colorectal cancer.
As Chair of the CALGB GI Committee I run a program that devises Phase II and III studies in GI cancers directed toward improving outcomes for GI cancer patients and understanding the biology of these diseases.
